The Origin of the Reaction of Harden and Young in Cell-free Alcoholic Fermentation* by Otto Meyerhof

چکیده

Since adenosine triphosphate (ATP)l transfers its labile phosphate to glucose, for every mole of triose phosphate oxidized 1 mole of inorganic phosphate is taken up by the fermenting system. Besides this uptake, an internal cycle exists for the second phosphate group, which is already present in the triose phosphate and after oxidation appears finally in the phosphopyruvic acid. This phosphate is also transferred to glucose or hexose monophosphate and reappears therefore in a new molecule of triose phosphate. This internal cycle does not add new phosphate to the system in the stationary state and can therefore be omitted from the discussion. However, another aspect of the reaction of Harden and Young is less obvious. In cell-free fermentation two phases follow each other. First there is a rapid phase, “phosphate period,” for which equation (1) applies;

Otto Fritz Meyerhof and the elucidation of the glycolytic pathway.

The elucidation of the glycolytic pathway, the process whereby glucose is converted into pyruvate and ATP, began in 1860 when Louis Pasteur observed that microorganisms were responsible for fermentation. Several years later, in 1897, Eduard Buchner made the significant discovery that cell-free extracts could carry out fermentation. The next important contribution was from Arthur Harden and Will...

A new coenzyme of alcoholic fermentation.

It is well known that cell-free alcoholic fermentation shows the HardenYoung effect: when sugar is fermented by dialyzed Lebedev maceration juice in the presence of orthophosphate and all known coenzymes, the rate of fermentation decreases markedly after the free phosphate is exhausted (cf. Meyerhof (1)). This phenomenon is due to the coupling between dehydrogenation of triose phosphate and bin...

Hexose Phosphates and Alcoholic Fermentation

The participation of phosphoric acid in alcoholic fermentation and in the biological utilization of carbohydrates in general, is one of the most important discoveries made in connection with the study of these systems. Numerous attempts have been made to correlate this discovery with the many observations regarding the carbohydrate-phosphates which are formed in such biologic processes, but opi...

Studies on Blood Glycolysis I. Effect of Arsenate

Meyerhof (8) has shown that arsenate stimulates the decomposition of carbohydrate and of hexosephosphate esters by the glycolytic enzyme of muscles. This observation brings muscle glycolysis into close relationship with fermentation of sugar by yeast or yeast products. Harden and Young (6) who discovered the stimulating action of the phosphate ion on the process of alcoholic fermentation were l...
